Cytotoxic Activity of Lymphocytes

Abstract
A simple, quantitatively reproducible assay of lymphocyte-mediated cytotoxicity which duplicates results achieved by microscopically counting residual target cells or by 51Cr release has been developed. The method is based on quantitation of the intracellular 86Rb incorporated by target cells in monolayer culture after exposure to sensitized lymphocytes. Factors of paramount importance in detecting cytotoxicity are degree of sensitization of the lymphocytes, antigenic density of the target cell, and use of optimal standardized geometric conditions for the assay. Elimination of plastic adherent cells is essential to demonstrate immunologic specificity of the attacking cells. Nonspecific destruction of syngeneic monolayers is avoided by this procedure.Keywords
