Abstract
The influence of metyrapone (SU4885; 2-methyl-l,2, bis-[3-pyridyl]-l-propanone), an adrenocortical blocking agent, upon aldosterone secretion was evaluated in 9 normal subjects and in 7 patients including 4 with Cushing''s syndrome, 2 with primary aldosteronism and 1 with hypopituitarism. Following equilibration on a constant diet restricted in Na, metyrapone was given orally in a dose of 750 mg at 4 hourly intervals for 2 consecutive days. Aldosterone secretion rates were measured on the day prior to and on the 2nd day of metyrapone administration. Despite the stimulus of salt deprivation, metyrapone was uniformly effective in reducing aldosterone secretion in all subjects. Concomitant ACTH stimulation in one normal subject failed to overcome the metyrapone induced block. In the majority of subjects, metyrapone administration was associated with a mild natriuresis.This publication has 1 reference indexed in Scilit:
