Abstract
To study a possible drug interaction between cimetidine (C) and sucralfate (S) [drugs used in the treatment of peptic ulcer], a crossover design was employed using 4 beagle dogs in each of the 3 groups: C alone, S alone, and C plus S. A 1 wk washout period elapsed between each treatment. C (300 mg, spiked with 3H material 9.9 .mu.Ci) was given P.O. [orally] by stomach catheter. S (1000 mg) was given as an intact tablet. To group 3, C was administered 1 h after S, dosing. In the presence of S. Ka of C decreased significantly from 3.99 .+-. 0.75 to 2.31 .+-. 0.19 (SD) l/h, and tmax [time to reach maximum concentration] increased significantly from 0.59 .+-. 0.12 to 1.00 .+-. 0.08 (SD) h. In the presence of C, Cmax [maximum concentration] and AUC (0-8) [area under the concentration time curve from from 0 to 8 h] of Al increased from 21.38 .+-. 4.78 to 29.25 .+-. 5.86 (SD) mcg/ml and from 132.3 .+-. 40.8 to 168.1 .+-. 22.0 (SD) (mcg/ml) h.This publication has 0 references indexed in Scilit:
