Abstract
The DNA sequences for 9 independent promoter mutants of a tyrosine tRNA gene, tyT of E. coli, are reported. The 9 mutations involve 6 transitions, 2 transversions and 1 deletion. They are located at 4 different sites in the first 30 base pairs preceding the start point of transcription. The changes found are: a T.cntdot.A to A.cntdot.T transition at position -8 (2 mutants); a T.cntdot.A to C.cntdot.G transition at position -13 (2 mutants); a T.cntdot.A to C.cntdot.G transition at position -16 (1 mutant); and a deletion of a G.cntdot.C base pair at position -26/27 (1 mutant). Four of the 5 different mutant tyrT promoters have alterations at positions that were probably expected from DNA sequence studies with other prokaryote promoters. One of these mutants (a G.cntdot.C deletion at position -26/27) occurs in a stretch of 8 consecutive G.cntdot.C base pairs which may be characteristic of stable RNA promoters.
