Short Term Glucose Homeostasis in Man: A Systems Dynamics Model

Abstract
A model of glucose homeostasis whose behavior resembles clinically observed behavior is subjected to a variety of glucose stimuli. This model is used to focus attention on the mechanism of insulin secretion as related to the duration of glucose infusion during the standard intravenous glucose test (0.5 gm/kg body weight). An important clinical experiment was suggested and performed—the duration of glucose infusion was increased from 3 minutes to 12 minutes. An initial peak of insulin is observed at 4 to 6 minutes after the beginning of the infusion. This is followed by a fall in insulin levels while the glucose continues to rise. Because of this experiment, the initial model of glucose homeostasis has undergone changes to include circulatory and transport dynamics and altered mechanisms of insulin release and liver clearance of both glucose and insulin.Keywords
