CORPUS LUTEUM STUDIES RECOVERY OF PREGNANDIOL FROM URINE

Abstract
An attempt was made to evaluate 2 methods for the detn. of pregnanediol excretion in the urine, by means of a combined method with which it is possible to recover both forms of pregnanediol from the same specimen of urine. The chief objection to detns. based only upon recovery of Na pregnanediol glucuronidate is the difficulty of preventing hydrolysis during collection of the urine. The chief objection to detns. based only upon the recovery of free pregnanediol is the occasional failure to convert all of the pregnanediol to the free form by incubation. The method based upon the detn. of Na pregnanediol glucuronidate has the advantage of a much heavier molecule which is an important factor in attaining accuracy since the yields are measured gravimetrically. The method based upon the recovery of free pregnanediol may have an advantage in the investigation of conditions where there is an infection in the urinary tract and where some hydrolysis occurs before the urine is voided. Where a reliable measure of pregnanediol excretion is desired (especially in hot weather) it seems best to supplement Venning''s procedure with a test for free pregnanediol as outlined in the combined method described above.
