Nitrate and ammonium as nitrogen nutrients for plant cell cultures

Abstract
A chemically defined 71V medium was designed in accordance with data from studies of NH4+ and NO3 requirements for biomass production of carrot cell suspension culture (CA-68 line). Under the described conditions, the optimum ratio of NH4+:NO3 was 20:80, and the optimum total nitrogen level was about 500 mg N per liter of medium. The observed changes in pH of the different inorganic media during the growth of the culture might be an important factor influencing the nitrogen metabolism of the cells.
