Abstract
A procedure is described which permits separate estimation of blood estrogen bound to protein, and estrogen not so bound. The procedure involved precipitation of the fresh blood sample with 10 vols. of ice-cold acetone, after which the supernatant fraction was separated and combined with the cold acetone-ether washings of the undenatured protein precipitate. Estrogen not bound to protein was present in this supernatant fraction, and could be removed by conventional procedures. The protein fraction was treated separately, by digestion with 0.1 [image] NaOH, followed by acidification and repeated extraction with ether at the centrifuge. Prepn. for assay of the estrogen fractions thus separated was described. Total blood estrogen values obtained by this method during the last trimester of pregnancy in the human agreed well with results by other procedures which measure total blood estrogen.Keywords
