The isolation of potato virus S from the leaves of potato plants using precipitation by polyethylene glycol

Abstract
A purified potato virus S was prepared using precipitation by the solution of 35% polyethylene glycol 4000 in the presence of an electrolyte. The mixture for precipitation of the potato virus S had to contain 11% polyethylene glycol and 0·25m NaCl. The S virus was extracted by 0·01m phosphate buffer, pH 7·5, from the precipitation separated by centrifugation. One part of the extract was further purified by means of differential centrifugation and the other by means of gel filtration on Sephadex G-100. The ultraviolet absorption measurements of both preparations showed that the differential centrifugation gave a purer preparation than the gel filtration. Byl připraven čistý preparát viru S bramboru precipitací 35% roztokem polyethylenglykolu 4000 za přítomnosti elektrolytu. K srážení viru S bylo zapotřebí, aby bylo ve směsi 11% polyethylenglykolu 4000 a 0,25m NaCl. Z odstředěné sraženiny byl virus S extrahován 0,01m fosfátovým pufrem o pH 7,5. Část extraktů byla dále čištěna diferenciální centrifugací a část gelovou filtrací přes Sephadex G-100. Proměření takto získaných preparátů viru S v UV oblasti spektra ukázalo, že diferenciální centrifugace skýtá čistší preparát než gelová filtrace. Приготовлен чистый препарат вируса S картофеля осаждением 35% раствором полиэтиленгликоля 4000 в присутетвии электролита. Для осаждения вируса S оказалось необходимым, чтобы в смеси было 11% полиэтиленгликоля 4000 и 0,25 M NaCl. Из отцентрифугированного преципитата был вирус S выэкетрагировах 0,01 M фосфатным руфером pH 7,5. Часть экстракта далее очищалась дифференциальным центрифугировабием а другая часть фильтрацией на Sephadex G-100. Измерения полученных препанатов вируса S в УФ области показали, что дифференциальное центрифугиронание дает облее чистый препарат, чем фильтрация на геле.Keywords
