Abstract
The amino acid sequences of the twenty peptides isolated from the 24-hour chymotryptic digest of performic acid-oxidized ribonuclease T1 [EC 2.7.7.26] were determined almost completely by various combinations of chemical and enzymatic methods. In addition, the location of the single tryptophan residue in the intact protein was elucidated by analysis' of the tryptophan-containing peptide derived from the 30-hour chymotryptic digest of heat-denatured ribonuclease T1. The combination of the information on the chymotryptic peptides with that on the tryptic peptides obtained earlier permitted us to deduce their unique alignment, including the location of the amide groups, hence the complete amino acid sequence of ribonuclease T1 (MW, 11, 085) H-Ala-Cys-Asp-Tyr-Thr-Cys-Gly-Ser-AspNH2-Cys-Tyr-Ser-Ser-Ser-Asp-Val-Ser- Thr-Ala-GluNHrAIa-Ala-Gly-Tyr-GluNH2, -Leu-His-Glu-Asp-Gly-Glu-Thr-Val- Gly-Ser-AspNH2-Ser-Tyr-Pro-His-Lys-Tyr-AspNHrAspNH2-Tyr-Glu-Gly-Phe- Asp-Phe-Ser-Val-Ser-Ser-Pro-Tyr-Tyr-Glu-Trp-Pro-Ile-Leu-Ser-Ser-Gly-Asp-Val- Tyr-Ser-Gly-Pro-Gly-Ser-Gly-AIa-Asp-Arg-Val-Val-Phe-AspNH2-Glu-AspNH2, - AspNH2-GluNH2--Leu-Ala-GIy-Val-Ile-Thr-His-Thr-GIy-Ala-Ser-GIy-AspNH2- AspNH2-Phe-Val-Glu-Cys-Thr-OH.Keywords
