An Apparatus for Use With Incremental Gradient Elution

Abstract
An apparatus for use with incremental gradient elution is described. It consists of a multi-solvent system fed from reservoirs connected to a multiport valve controlled by an electronic programmer. A modified mixing vessel is also described and its performance characteristics given. The design of a pulse damper that will not interfere with the gradient elution process is included together with results demonstrating its efficiency. The effect of pump dead volume on solvent concentration profiles is determined and a method of improving pump performance for gradient elution is given. The extent to which the moving wire detector contributes to band width is examined, and an example of the use of the apparatus in the separation of a solute mixture containing substances of widely differing polarities is included.
