An Optimal Assay System for Subcellular Determination of De Novo Fatty Acid Synthesis in Human Adipose Tissue

Abstract
An optimal assay system is described, which determines the overall enzyme capacity for de novo fatty acid synthesis in human adipose tissue by measuring the simultaneous incorporation of acetyl units from acetate and citrate. Incorporating activity was almost exlusively recovered in the cytoplasmic fraction of the fat cell. Several assay conditions were crucial. Practically linear incorporations with time and enzyme concentration were attained. The main products of the system were myristic and palmitic acids. Acetate and citrate incorporations were of the same magnitude and demonstrated similar distribution patterns.
