Abstract
Incubation of fresh conjunctiva in media containing Na4EDTA (40 mM/ml) or elastase (30 units/ml) and fixed tissue in collagenase (150 units/ml) was found to sufficiently loosen the epithelium from its underlying lamina propria to allow mechanical separation of the tissues. It was found that such treatment yields intact epithelia uncontaminated by connective tissue as evaluated at the light-microscopic level. Flatmounting the isolated epithelia upside down renders the proliferative cell population, located in the basal layer, accessible to autoradiographic and histochemical analysis.Keywords
