Blood levels of rifampicin, desacetylrifampicin and isoniazid during combined therapy

Abstract
Some pharmacokinetic properties of rifampicin and isoniazid, alone and in combination therapy, were studied by determination of the bloodlevels of these antibiotics and of the metabolite desacetylrifampicin, in 12 tuberculosis patients on a programmed therapeutic schedule. The results expressed as the apparent eliminationrate constant (kapp and as the area under the serum concentration curve (AUC) were statistically analysed. The following conclusions were drawn: I, induction of metabolism of rifampicin occurs; 2, the dose (600 versus 1200 mg) does not affect this induction; 3, isoniazid has no influence on the induction but it affects rifampicin bloodlevels unfavourably; 4, increase of k of rifampicin is associated with a decrease of desacetylrifampicin bloodlevels, 5, isoniazid bloodlevels are not affected by concomitant rifampicin therapy.Keywords
