Measurement of Glucuronic Acid Metabolites by High-Resolution Gas Chromatography

Abstract
An analytical procedure is presented that determines major metabolites of the D-glucuronic acid pathway, specifically D-glucaric acid, D-glucuronic acid, L-gulonic acid, and xylitol. A capillary gas chromatographic system with flame ionization detection is used to simultaneously determine the silylated (trimethylsilyl) derivatives of each metabolite. Derivatives are stable, and the standard curves for each metabolite, glucaric acid, glucuronic acid, gulonic acid, and xylitol, are linear up to 30, 800, 10, and 30 nmol, respectively. The specimen type is urine and is therefore noninvasive to humans. This analytical technique can be used to assess exposure to chemicals that have been shown to increase urinary concentrations of these substances.
