Antitumor Activity of 5-Bis(2-Chloroethyl) Amino Uracil Against Sarcoma 180 and Ehrlich Carcinoma

Abstract
The acute LD50 in Swiss mice of 5-bis(2-chloroethyl) amino uracil (U-8344) and chlorambucil (CB 1348) was 3.5 mg/kg and 63.5 mg/kg respectively. 0.8 mg/kg of U-8344 given for 3 days increased the survival of Ehrlich carcinoma ascitic mice from 16 to 43 days (P =<0.01); chlorambucil (10 mg/kg/day) from 16 to 19 days (P = 0.07). The total packed tumor cell volume of Sarcoma 180 or Ehrlich carcinoma ascitic mice is significantly reduced (P =< 0.01) when the mice are treated with either 0.33 mg/kg/day of U-8344 or 10 mg/kg/day of CB 1348. 0.8 mg/kg/day of U-8344 produced a 69-83% inhibition of S-180 (solid) which is significant (P = 0.01). 40% inhibition of established Sarcoma 180 was obtained with 1.6 mg/kg of U-8344 (P = 0.02).Keywords
