Abstract
Methods are described for the immobilization of single-stranded DNA by trapping in gels of cellulose acetate or agar. The immobilized DNA retains the ability to form specific hybrids with complementary RNA. This provides a general method for the isolation of RNA molecules having sequence homology with DNA molecules from any given source. The specificity and kinetics of the hybridization reaction are examined, and the method is illustrated by a study of the rate of synthesis of complementary RNA in Proteus vulgaris.Keywords
