Isozyme Genetics and Linkage in Tepary Bean, Phaseolus acutifolius A. Gray

Abstract
The inheritance, genetics, and linkage of 12 variable isozymes from eight enzyme systems in Phaseolus acutifollus A. Gray (tepary bean) were determined using F2 progenies. The allozymes were encoded by codominant alleles of nine simply inherited loci, and four pairs of linked loci were detected. Moderate linkage was detected between loci Fdh-1 and 6pgd-2, Gpi-c1 and 6pgd-2, and Aco-2 and Idh-x. Adh-1 and Gpi-c2 were tightly linked; no recombinants were detected between these loci. Pgm-2 and Skdh-1 segregated independently of the others. The proposed quaternary structures of the various enzymes, with the exception of 6PGD-2, conformed to previous reports. ADH-1, FDH-1, IDH-4, GPI-1, and GPI-3 behave as dimers, and ACO-2 and PGM-2 behave as monomers. The formation of an active hybrid isozyme from the polypeptides of Gpi-c1 and Gpi-c2 and the independent segregation of these two loci provide evidence of a Gpi duplication in tepary bean.
