Abstract
Methods are given for the elimination of background material in estimating numbers in low density populations of entozoic amoebae. Rice starch is purified by digestion with trypsin and unusable material is eliminated by differential centrifugation and sedimentation. Excess starch present at the time of counting is rendered optically transparent by adding 10% formalin and heating for 10 min. at 70[degree]C. Hydrolysis of the starch with HC1 was not necessary but introduced no error in the counts. Keywords
