Metabolic Activities of Sloughed Cells from the Jejunal Mucosa of Dogs with Thiry-Vella Loops.

Abstract
The metabolic activity of sloughed jejunal mucosal cells was studied by 3 different methods. Differential staining with Nigrosin showed that those cells in the crypts of the villus have intact cell membranes and do not take up the dye while those cells at the tip of the villus and those which are extruded are permeable to the dye. Labelled acetate was incorporated into non-saponifiable lipid by the intact jejunal epithelium while the sloughed epithelial cells were unable to accomplish this synthesis. The particulate fraction of the succus entericus, however, contained 95% of the total sucrase and alkaline phosphatase activity present in the secretion. The sucrase activity was stable during incubation with the intestinal secretion. A dependable in vivo method for determining sloughing rate based on the stability of sucrase and the use of a simple flushing procedure is described.
