Abstract
Permeation chromatography on controlled pore glass (CPG) was compared with sucrose density gradient centrifugation for the purification of 15 widely differing plant viruses. CPG chromatography gave excellent recovery of nearly all the viruses as evaluated by E260 [extinction coefficient], serological titer and infectivity assay and proved as good as or better than sucrose density gradient centrifugation. Both methods gave similar separations of virus from host [plant] material and acceptably pure virus preparations. CPG columns accepted virus in high or low concentration; non-specific adsorption was not observed using CPG pre-treated with poly(ethylene glycol) of MW 20,000.
