On the Cellular Fixation of Cytophilic Antibody

Abstract
The cytophilic antibody receptor activity of rabbit cells in vitro was investigated. Different types of rabbit cells fix rabbit cytophilic antibody. Spleen cells from other animal species also fix rabbit cytophilic antibody. The proteolytic enzymes trypsin, papain and chymotrypsin, 8 [image] urea and 5% phenol rapidly destroy antibody receptor activity of rabbit spleen cells. Organic lipophilic solvents, Tween 80 and lipase also reduce, although not to the same extent, the capacity of cells to take up cytophilic antibody. Pretreatment of cells with SH blocking agents and S-S reducing compounds interferes with antibody fixation. The results of chemical, physical treatment and inhibition tests suggest heat labile non-specific protein, possibly lipoprotein sites which are concerned with antibody fixation on spleen cells. Mitochondria and microsomal particles, but not the nuclear fraction, can bind cytophilic antibody.Keywords
