Abstract
[beta]-Thymonucleic acid can best be prepared from nucleus-rich material (fish sperm) by 4-day treatment at weakly alkaline reaction with pancreatin, the nucleogelase of which depolymerizes the product to the easily-purified [beta]-form. With nucleus-poor material (tissue) it was better to effect a preliminary purification by NaOH hydrolysis and alcohol precipitation before depolymerization with pancreatin. Operational details are given.
