QUANTITATIVE-DETERMINATION OF TOBRAMYCIN WITH THE AID OF HIGH-PRESSURE LIQUID-CHROMATOGRAPHY

Abstract
Separation of the main components of the nebramycin complex of apramycin, kanamycin B and tobramycin was achieved by obtaining their 2,4-dinitrophenyl derivatives. A SR 8000 chromatograph and a Spherisorb C6 column (4.6 .times. 250 mm) with a granule size of 5 .mu.m were used with a buffered mobile phase of pH 7, a flow rate of 1 ml/min, a temperature of 35.degree. C and detection at .lambda. 350 nm. Quantitative determination of the tobramycin purity level was performed with an equation developed for the given conditions. The purity levels of the reference tobramycin base and its sulfate were estimated.
