A Solid-Phase Radioimmunoassay for a Thermostable Adrenal-Specific Antigen

Abstract
A radioimmunoassay for the detection of a thermostable antigen of bovine adrenal was described. Antibodies labelled with 125I were ‘purified’ by absorption with nonadrenal antigens of bovine origin and by recovering antibodies adsorbed to plastic tubes coated by adrenal antigen. The test was performed by counting radioactivity on antigen coated tubes after their reaction with radio-labelled purified antibodies. The antigen was quantitated on the basis of the reduction of the radioactivity which resulted from the neutralization of antibodies. The sensitivity of the test was satisfactory in that 15 ng of antigen/ml of serum could be detected. In preliminary experiments performed on 5 rabbits, the antigen in concentrations up to 180 ng/ml was detected within 24 h after injection of 0.5–1.0 g of a homogenate of bovine adrenal.Keywords
