PROTEIN SYNTHESIZING SYSTEM FROM WHEAT-GERM - EFFICIENT TRANSLATION OF SYNTHETIC AND NATURAL MESSAGES

Abstract
The optimum conditions for translation of eukaryotic, prokaryotic and synthetic templates in wheat germ cell-free extract were determined. Translation of eukaryotic messages (BMV [Brome mosaic virus] RNA and TMV [tobacco mosaic virus] RNA) was optimum at the same concentrations of K+, Mg2+, N-2-hydroxyethylpiperazin-N''-2 ethanesulfonic acid and spermine. In optimal conditions the efficiency of translation was high, being equal to 220 pmol of leucine incorporated/.mu.g template for BMV RNA and 280 pmol of leucine for TMV RNA. Prokaryotic template (Q.BETA. [Q.BETA. phage] RNA) was translated under different ionic conditions. Translation of the synthetic template poly(U) was optimum at higher concentrations of K+ and Mg2+ than those optimal for natural template translation. Efficient translation of natural and synthetic templates depends on complete removal of inhibitors found in the wheat germ cell-free extract.
