In Vivo Action of a New Lipotropic Fraction of the Pancreas

Abstract
A new lipotropic fraction of beef pancreas prepd. by the method of Bosshardt, Ciereszko and Barnes was tested in dogs as follows. Of 19 dogs reported, 17 were depancreatized (and maintained with insulin) and 2 served as controls for the diet which consisted of casein 20%, lard 10%, carbohydrate 66%, salt mixture 4% and a vit. mixture considered adequate for the dog. Of the 17 depancreatized dogs, 7 served as controls, 5 received 60 mg. per day of the extract and 5 received 60 nag. of extract which had been boiled for 3 min. Liver biopsies done under local anesthesia about one month after pancreatectomy and after 3-4 weeks on the diet showed a mean fat content for the intact control animals (not depancreatized) of 4%, for the depancreatized control animals of 14.6 [plus or minus] 2.3%, for the group receiving the extract 6.4 [plus or minus] 1.2%, and for the group receiving the boiled extract 16.8 [plus or minus] 4.4%. Animals which stopped eating tended to develop fatty livers despite the extract. This data indicated that the extract exerted a marked lipotropic effect in the depancreatized dog, that its action was largely destroyed by boiling and it appeared probable that it acted in the body as an enzyme.
