The Estimation of Cytochrome C Oxidase in Animal Tissues

Abstract
A method for the precise estimation of the cytochrome oxidase in animal tissues was described. The addition of 1-phenylsemi-carbazide to the system in vitro containing hydroquinone as a substrate prevented the accumulation of inhibitory reaction products. Semicarbazide which was used formerly for this purpose was unsuitable since a gas, probably nitrogen, was formed when it reacted with the oxidation products of hydroquinone. The effect of the phosphate buffer, and of other salts, upon the activity of the enzyme was described. It is important, for an estimation of cytochrome oxidase, that not only should the cytochrome concn. be adequate, but also the concns. of buffer and other electrolytes should be controlled.
