Influence of Genotype and Environment on Growth of Barley Embryos in vitro

Abstract
Isolated embryos of three contrasting barley genotypes were cultured in vitro on a range of media comprising 16 combinations of sucrose (3–12 per cent) and 2, 4-D (0–8 mg 1−1) concentration. Cultures were incubated at a range of temperatures from 5 to 25°C and were examined after 21 days when shoot lengths as well as fresh and dry weights were recorded. The relative influence on growth of increasing concentrations of sucrose and 2, 4-D was investigated, as was their interaction with the incubation temperature. The genotypes were found to differ markedly in their response to these two media components, with each parameter of growth differentially affected. Results are discussed in relation to the known dormancy characteristics of these genotypes.Keywords
