An Improved Method for Isolating Viable Human Leukocytes from Peripheral Blood

Abstract
In an effort to isolate human leukocytes as free as possible of erythrocytes, a two-stage method has been studied. The initial step, sedimentation of the erythrocytes with Dextran results in a diminution of RBC:WBC ratio from the 1000:1 of peripheral blood, to 2.34:1. After sedimentation in isotonic saline, the RBC:WBC ratio is decreased to 0.16:1. This method should prove valuable in preparing leukocytes for immunologic studies.Keywords
