Toxicity of Chronic Dichloroacetate

Abstract
To the Editor: Early chronic, oral-toxicity studies of the di-isopropylammonium salt of dichloroacetate in dogs revealed no noteworthy histologic or clinical evidence of drug toxicity.1 Since publication of our initial report on the effects of sodium dichloroacetate in patients with diabetes mellitus or hyperlipoproteinemia,2 however, subsequent investigations have disclosed adverse drug-related effects in animals and in at least one human subject that seriously limit any therapeutic potential of this agent.Oral administration of dichloroacetate for 90 consecutive days to rats in doses of 125, 500 and 2000 mg per kilogram of body weight3 has been found to be associated with . . .
