Enhancement of Mammalian Cell Sensitivity to Hyperthermia by pH Alteration

Abstract
The effect of pH on the growth rate and reproductive capacity of Chinese hamster cells was examined at 37.degree. and 42.degree. C. At 37.degree. C, the population doubling time did not vary between pH 7.4 and 7.0, but was prolonged at lower pH. Cell viability was not affected from pH 7.4-6.7. The effect of 42.degree. C on the viability of cells was influenced by pH. Four hours of heat treatment resulted in a 10 fold decrease in survival at pH 7.0 and a 500 fold decrease in survival at pH 6.7, relative to pH 7.4. These results indicate that pH variations may play a role in determining sensitivity to hyperthermia. This pH effect may be significant under conditions which stimulate metabolic acidification.
