Free Intracellular Amino Acid of a Glutamate-Forming Bacterium, Brevibacterium flavum No. 2247

Abstract
Amino acid compositions of free intracellular pool and external medium of Brevibacteriwn flavum No. 2247 grown in biotin poor and -rich media were analysed micro-biologically. Of 18 amino acids, the major components of internal amino acid pool were glutamic acid, alanine, valine, lysine, and serine; and the major components of external medium were glutamic acid, aspartic acid, and alanine under biotin-poor condition, and alanine alone under biotin-rich condition. Under biotin-poor condition, amino acid contents of internal pool were much less than those under biotin-rich condition and all the amino acids decreased strikingly on washing with a buffer, while under the latter condition acidic and basic amino acids did not decrease significantly on washing. These behaviors seem to be correlated with the extracellular accumulation of amino acids in B. flavum.Keywords
