THE OXIDATION OF ACETATE BY MICROCOCCUS LYSODEIKTICUS

Abstract
Incorporation of isotope from the substrate into non- isotoxic carriers suspected of being intermediates in the metabolism of the substrate is not sufficient evidence to determine whether the carrier is or is not an intermediate. The degree of equilibration of the intermediates with each other, with the substrate and with the respiratory CO2 are important. During the oxidation of acetate, equilibration does not occur with suspected intermediates of the tricarboxylic acid cycle used as carriers. On the other hand, intermediates isolated from large scale expts., in which the intermediates are presumably present as substrate-enzyme complexes, show complete equilibrations with each other, acetate and respiratory CO2. It is concluded that the tricarboxylic acid cycle is of quantitative importance in M. lysodeikticus.Keywords
