A rigorous analysis of the methodological aspects of enzyme localization procedures which demonstrates both in principle and in practice the importance of the following criteria: (1) zero order breakdown of substrate, (2) first order capture reaction of adequately high velocity constant, and (3) non-diffusibility of visualizing substance resulting from low solubility and high substantivity. The development of a method for esterases which appears to conform with these criteria is described.
