Analysis of phenylthiocarbamyl-amino acids at pico-mole level by high performance liquid chromatography and application to plant materials

Abstract
Pre-column derivatization with DNS (BAYER et al. 1976), PTH (BLACK and COON 1982; KOBAYASHI and TARR 1986), OPA (LINDROTH and MOPPER 1979: WINSPEAR and OAKS 1983), or PTC (BIDLINGMEYER et al. 1984; KOBAYASHI and TARR 1986; KUWANO et al. 1987) enables the separation of ammo acids by reversed-phase HPLC. Analysis of amino acids with these methods was efficient and accurate. However, one of the drawbacks of the methods using OPA is the lack of derivative stability and of reactivity with several amino acids, such as proline (LINDROTH and MOPPER 1979; WINSPEAR and OAKS 1983). Interference from reagent peaks is also a major problem with DNS (BAYER et al. 1976).Keywords
