Trp284 of the Human δ Opioid Receptor is Required for SNC121 Binding but Not Binding of pCl-DPDPE, Deltorphin II or Naltrindole

Abstract
In this report we show that recognition of the nonpeptidic ligand SNC121 by the human δ opioid receptor depends on the Trp284 residue of the third extracellular loop. The binding affinities of other opioid ligands including the antagonist naltrindole and the selective δ receptor peptide agonists [4′-ClPhe4]DPDPE and deltorphin II, are not affected by this mutation consistent with its specificity for SNC121. Thus the Trp284 residue of the human δ opioid receptor is part of the recognition site used by SNC121 but not by other opioid ligands.Keywords
