Susceptibility of Chromatin to Nuclease Digestion in Dormant and Proliferating Meristems of Allium cepa Roots

Abstract
Digestion of chromatin with microccal nuclease has been carried out on intact cells of Allium cepa L. meristems previousiy made permeable by ethanol-acetone treatment. Digestion is more rapid in proliferating meristems compared with dormant ones, as measured by microdensitometry after Feulgen staining. The greater accessibility of chromatin in the nuclei of proliferating meristems to enzyme is related to the lesser amount of dense chromatin they contain and may be linked to their increased replication and transcription processes.
