Abstract
No staining methods are involved. The pancreas is placed on a black plate, in a drop of saline, and flattened to a thickness of 180 microns by a transparent glass plate. The islets stand out as white spots on a dark acinar background; there is histological evidence that the white spots are identical with islet tissue. The prepn. is photographed, and the areas represented by islet and acinar tissue measured by a projection method. Volume is estimated by multiplying area by thickness: 180 microns for acinar tissue, and 47 microns (avg.) for islet tissue. The islets may be counted directly.
