Abstract
Countercurrent distr. of prepns. of growth hormone in the system 0.005 [image] p-toluenesulfonic acid: 2-butanol produced at least one major and varying amts. of a minor component. The best prepn., supplied by Prof. Li and stated to be homogeneous by electrophoresis, chromatographic column and end-group analysis, contained only 3% of the minor component. Growth-promoting activity was present in the major component, the distr. curve of which agreed well with the theory for a single substance. With 0.005 [image] p-toluenesulfonic acid, the major component had a partition coefficient of about 10. A system with 0.0022 [image] p-toluenesulfonic acid was used in an attempt to obtain a more favorable partition coefficient for the study of homogeneity of the major component. In this case the distr. curve of the major component was wider than the theoretical curve. Possible reasons for this deviation from the theoretical are discussed. A comparison between the distr. curve of a preparation of growth hormone and its electrophoresis pattern is presented.
