Data-Pair Technique, a New Approach to Quantitative Thin-Layer Chromatography

Abstract
The critical parameters for the application of in situ UV reflectance spectroscopy to TLC have been studied. The principle of “data-pair” formation has been developed. Reference and standard solutions are spotted in such a way as to give a pair for each concentration, with the two spots being about one half plate width apart. The average of four readings per spot (two forward and two backward scans in the direction of chromatography) is then taken. With this approach one can significantly reduce systematic errors due to chromatographic parameters which tend to influence reflectance values as a function of varying Rf values. The statistical validity of such a spot application and evaluation scheme is demonstrated. With the data-pair technique one can carry out three analyses per plate with a relative standard deviation of about ± 1.5%.Keywords
