Abstract
Various amts. of pamaquine naphthoate were administered to 36 paretics who received mosquito inoculations of Plasmodium vivax and P. falciparum. Daily doses of either 160 or 180 mg. on the day before, day of, and for 5 or 6 days following P. vivax injn. have prevented or postponed primary attacks for months. Similar doses for as short as 3 days after inoc. with P. falci-parum seem to have prevented disease. Two acute hemolytic anemias were noted among the 11 Negroes. Detailed data concerned with the toxic reactions, plasma pamaquine, and methemoglobin levels are tabulated and discussed. All patients developed methemoglobinemia. Pamaquine is both rapidly absorbed and excreted, but the plasma levels appear independent of mg./kg. dosage. Keywords
