THE INITIATION OF VIRAL PROTEIN SYNTHESIS IN E. COLI EXTRACTS

Abstract
Studies suggesting that the codon for N-for mylmethionine is also read as an internal codon imply that some codons for N-formylmethionine may not initiate synthesis. A single triplet may signal the initiation of protein synthesis. The secondary structure in many regions of the RNA may restrict interaction with ribosomes. The synthesis of viral proteins in Escherichia coli extracts depends on N-formylmethionyl-sRNA or a formyl donor. At low Mg concentrations, synthesis completely depend on formylation. At about 0.009 [image] Mg where synthesis was maximal, stimulation ranging from 10-28-fold were obtained by the addition of a formyl donor.
