Abstract
A histochemical method for the localization of amylase in tissue sections is described. A thin film of starch is exposed to a tissue section and, after incubation, is stained by the periodic acid-Schiff reaction. Clear areas are then observed in the film due to hydrolysis of starch by the amylase present in the tissue. Comparison of the unstained pattern in the starch film with the corresponding tissue section reveals the sites of amylase activity in the tissue.
