THE USE OF THE GEVAERT NUC-307 NUCLEAR EMULSION FOR RADIOAUTOGRAPHY AT THE ELECTRON MICROSCOPE LEVEL

Abstract
The fine-grain Gevaert NUC-307 emulsion was investigated for use in radioautography. First, an attempt was made to develop grains visible in the light microscope in order to compare the sensitivity and the background fog of this emulsion with that of other emulsions and thus obtain the data required to devise an adequate set of conditions for work with the electron microscope. A good method for electron microscopic radioautography was to dip EM grids attached to glass slides in undiluted NUC-307 emulsion warmed to 40°C. The exposed preparations were developed for 5 minutes in D-170 at 18°C. The silver grains were found to be smaller and, therefore, to allow a better resolution than those of Ilford L4 emulsion. The high resolution as well as the high sensitivity of the NUC-307 emulsion make it well suited for electron microscopic radioautographs.Keywords
