Abstract
Sensitivity of the barley-endosperm bioassay for gibberellins exhibits considerable variability. In part this was shown to be due to the variety used and the location in which seed was grown. The age of seed affected response, and sensitivity also varied from season to season. Time of incubation was of special significance and at least 30 h at 30 °C is recommended. Significant losses of reducing sugar during certain longer periods of incubation were encountered with some gibberellins. However, the period when loss occurred, or the particular gibberellin with which this was found, varied between different tests. Response to gibberellins subjected to thin-layer electrophoresis or chromatography may be improved if scrapings are subjected to vacuum- or freeze-drying before bioassay.
