Abstract
Extracts prepared from urine by 3 different methods were tested for the presence of insulin by their ability to lower the blood sugar of a fasted rabbit. Methods that involve the direct inj. of urine into the rabbit or alcoholic extraction of the urine are unsatisfactory. Precipitation of the insulin from the urine at pH 5.0 by half saturation with (NH4)2SO4 followed by elution of the insulin from the precipitate with a borate buffer of pH 8.0 gave good results. Using this technique it was possible to detect the presence of 0.25 units insulin per 100ml. when this had been added in vitro, but no insulin could be detected in normal urine. If present it must be there in amts. less than 0.25 units per 100ml.
