Identification of Inorganic Particulates in a Single Histologic Section Using Both Light Microscopy and X-Ray Microprobe Analysis

Abstract
This communication describes a technique which allows an investigator to examine the same paraffin section by light microscopy (LM) and by scanning electron microscopy combined with energy dispersive x-ray microanalysis (SEM/EDX). After careful examination by LM, the section is coated with Krylon® clear acrylic spray-on plastic, allowed to dry, removed from a glass slide and placed on a spectroscopically pure ultrasmooth carbon stub. After removal of the plastic with xylene the section can be examined by SEM/EDX. Using previously recorded light micrographs the investigator can identify with precision polarizable and non-polarizable foreign bodies, granulomas, etc., and determine the elemental composition of these foci by EDX.
