THE AMYLASE OF CLOSTRIDIUM ACETOBUTYLICUM

Abstract
Substances other than starch, such as fructose, mannose, mannitol,cellobiose and beta-limit dextrin stimulated the production of the adaptive enzyme, amylase. The enzyme was freed of maltose by treatment with acetone or acetone and ammonium sulfate and stored for 72 hrs. in the refrigerator. The maltase-free amylase digested amylopectin up to 80% maltose formation. A crude amylase prepn. digested beta-limit dextrin to 100% ferment-able products. An analysis of amylase activity indicated that enzymatic hydrolysis of starch consisted of 3 consecutive first-order reactions. Each reaction was substantially completed before the next reaction commenced. 97% of the theoretical max. ethanol yield was obtained with this amylase as a saccharifying agent.
