Production of Mold Amylases in Submerged Culture

Abstract
More than 350 spp. of fungi, including cultures of Rhizopus, Mucor, Penicillium, Aspergillus and Monilia, were investigated for their amylase-producing ability when grown under submerged aerobic conditions. While most of the organisms showed limited ability to produce amylase under these conditions, a few spp. of Aspergillus gave excellent enzyme production. Substantial amts. of dextrinizing enzyme were produced by cultures of A. oryzae, A. wentii, A. alliaceus and A. niger. In addition to dextrinizing enzyme, appreciable amts. of saccharifying enzyme were elaborated by A. niger. The more actively saccharifying prepns. were characterized by high maltase values. Culture liquors from A. niger, NRRL-337, were satisfactory replacements for distillers'' malt in the alcoholic fermentation of corn. The culture medium consisted of thin stillage supplemented with 2% of corn meal and 0.5% of CaCO3. Other protein and carbohydrate substrate materials gave smaller but satisfactory enzyme yields. Cultures were incubated at 28-30[degree]C under continuous aeration for 3-5 days.Keywords
